When Speeduino is off it will sends its injector outputs to ground.

Make sure you do not have your injectors powered when ECU is off.

It turns out that the injector can fill the cylinder if I did not immediately start the engine?

Well you have messed up the wiring or speeduino assembly, if that happens.

I hope I understand your question. Speeduino injector outputs will show +12V when off, as the voltage passes to the output through the Injector LED and LED resistor. No problem, and the injectors will not operate, as they need Speeduino to ground the powered injectors in order to work.
